Occasional Low-Protein Days
DeLauer presents this protocol as a practical application of the protein sensitivity concept. He reasons that if constant protein intake might blunt the muscle-building signal, occasional low-protein days could act as a 'reset.' During the low day, mTOR is suppressed, autophagy ramps up, and the body may upregulate amino acid recycling to spare muscle. When high protein is resumed, the surge in mTOR, IGF-1, and testosterone responsiveness drives a super-compensatory anabolic rebound, leading to greater net muscle protein synthesis than if protein was kept uniformly high. He supports this with the bolus-study evidence that longer gaps between protein feedings increase aggregate muscle protein synthesis, and with intermittent fasting research showing muscle preservation despite reduced calories and lower anabolic hormones. He personally implements this by dropping from 200-300g down to 50g on select days, noting it also feels good digestively.
Reducing protein intake decreases circulating amino acids and insulin, which lowers mTORC1 activity and stimulates autophagy. This clears misfolded proteins and damaged organelles, making cells more efficient. Simultaneously, the body increases proteostatic mechanisms to recycle amino acids, preventing muscle breakdown. Upon reintroduction of a large protein bolus, the combination of primed mTOR signaling and heightened amino acid transporter expression leads to a potentiated anabolic response, similar to the 'anabolic rebound' observed after short-term fasting. DeLauer also speculates that short-term suppression of testosterone and IGF-1 during low-protein periods may increase receptor sensitivity, so the same hormonal signal triggers a bigger effect when protein returns.
He says, 'I'm eating two to 300 grams of protein a day, which means that maybe I'm getting very desensitized to that. So, I've occasionally said, "Okay, I'm just going to drop my protein down to like 50 grams today and I'm not going to waste away and I'm going to do that to sort of cleanse the system a little bit."'
